Overview
Semax is a synthetic peptide, also developed in Russia at the same institute as Selank, and it is based on a short fragment of the natural hormone ACTH with a stabilizing extension added. It is studied as a neuroprotective nootropic, meaning a compound examined both for supporting cognition and for protecting brain tissue. Where Selank is framed around calm, Semax is framed around focus and mental drive, which is why the two are so often discussed and studied together. It has been used clinically in Russia as a nasal spray for conditions involving the brain and circulation. It sits in the cognitive category beside Selank.
How it works
Its most studied mechanism is a rapid increase in brain derived neurotrophic factor, or BDNF, a protein that supports the survival of existing neurons and encourages the growth of new connections between them. BDNF is sometimes described as fertilizer for the brain, and low levels are linked in research to cognitive decline, so a compound that raises it draws strong interest. Studies report that Semax also increases a related factor called NGF and switches on the downstream signaling that these growth factors use. Beyond the neurotrophins, research describes effects on the dopamine and serotonin systems, which may help explain why it is associated in studies with drive and task initiation rather than calm. These actions point toward an effect on neuroplasticity, the brain’s ability to adapt, rather than simple short term stimulation.
What researchers study
In animal models, Semax is examined for learning, memory, and attention, and for protecting brain tissue in models of stroke and low oxygen, which is where its neuroprotective framing comes from. A frequently cited 2006 study reported that a single intranasal dose produced a rapid rise in BDNF in a key brain region within hours. Researchers study both the fast acting effects on neurochemistry and the longer range implications for plasticity and recovery. Like Selank, Semax is a registered medicine in Russia but is not FDA approved, and much of its literature is concentrated in Russian research. The combination of a clear molecular handle in BDNF and a clinical history abroad keeps it an actively studied nootropic.
